What Is An API?

API refers to the pharmaceutical ingredients used to produce various kinds of preparations and is the active ingredient in the preparations. It is prepared through chemical synthesis, plant extraction or biotechnology to form powder, crystals, extracts and other forms, but the patient cannot take it directly; it generally needs to have excipients added, processing and other steps to be made into a pharmaceutical preparation that can be used directly.

APIs are the basis of drug development and production, and their quality directly affects the quality of the preparation, so countries have developed strict national pharmacopeia standards and quality control methods.

What Is A Pharmaceutical Intermediate?

According to the EU REACH regulation, an intermediate is defined as "a substance that is manufactured, consumed or used in a chemical process for the purpose of transforming the substance into another substance." Such substances do not normally appear in the final product, except as impurities. In the chemical industry, "intermediates" usually refer to semi-finished products that are generated during organic synthesis and are intermediate in the production of the final product. For example, in drug synthesis, intermediates are bridges between different chemical reactions and are used for further processing into drugs.

The Core Difference between APIs and Intermediates

In drug manufacturing, the core difference between an API and an intermediate is their pharmacological activity and end use.

APIs

APIs are active ingredients that have clear pharmacological activity or other direct effects in the diagnosis, treatment, symptomatic relief, control or prevention of disease, or are capable of affecting the function or structure of the human body, and can be used directly in the final clinical formulation.

Pharmaceutical Intermediates

Intermediates are intermediate products in the production of an API that do not have pharmacological activity and can only be used to further synthesize the next product, which is a key pre-process in the production of an API.

APIs and intermediates play different roles in the pharmaceutical industry chain, and together they contribute to the efficient, safe and high-quality production of drugs.

Case 1: Ubrogepant Supplied for Early-Phase Migraine Receptor Binding Research

A neuroscience research team at a pharmaceutical R&D center in the UK sourced ubrogepant from Alfa Chemistry to conduct non-clinical receptor binding assays targeting calcitonin gene-related peptide (CGRP) pathways. The compound was used to benchmark in vitro activity profiles of novel CGRP antagonists under development for migraine therapy. Alfa Chemistry provided non-GMP grade ubrogepant [Catalog Number YM00001] with complete HPLC purity profile (≥99%) and structural data (¹H NMR, MS).

"Using ubrogepant as a reference compound enabled us to validate our assay system. Alfa Chemistry's documentation and consistency made it suitable for scientific benchmarking purposes."
